Optimization of physicochemical parameters for biological synthesis of gold nanoparticles by Pseudomonas putida LUA 15.1

Abstract: Nanoparticles are synthesized through various ways such as physical, chemical and biological. Biological method of nanoparticles synthesis is eco-friendly approach, which yield no risk to the environment. Gold nanoparticles have wide area of applications especially in the field of medicine. This study aims to investigate the optimum physicochemical parameters for maximum synthesis of gold nanoparticles by Pseudomonas putida LUA 15.1 using hydro chloroauric acid as substrate. Different physicochemical parameters such as incubation time, temperature, pH, sources of nitrogen like peptone, beef extract, concentrations of NaCl and HAuCl4 and inoculum size have been optimized. The change in colour from pale yellow to wine red indicated the gold nanoparticles synthesis. Maximum gold nanoparticles synthesis achieved at 36˚C, pH 8.0, after 48h of incubation when 2.0mM hydro chloroauric acid was used, whereas 5.0g/l peptone, 4.5g/l beef extract and 5.0g/l of NaCl found to be optimum for maximum gold nanoparticles synthesis. Biosynthesized gold nanoparticles were further characterized using UV–vis spectroscopy which showed maximum absorbance between 520- 540nm, Fourier transform infrared spectroscopy (FTIR) depicted two bands at 3700cm-1 and 1500cm-1 which revealed the presence of amines, amides and carbonyl stretch in protein, Dynamic Light Scattering (DLS) graph revealed that, diameter of maximum gold nanoparticles fall in the range of 50nm.
